I trying to create a PEGylated liposome through the post-insertion method. This method uses pre-formed liposomes and incubates them with peptides conjugated on micelles to transfer the peptide from the micelle to the liposome. However, all methods to create micelles are very similar to the techniques used to create liposomes. How can I ensure that I am conjugating peptides on to micelles and not liposomes in the first step?
You need to calculate the approximate parameter of the critical packaging of your intended micelle. Maybe your substance does not form ordinary micelles, but only forms liposomes.

Generally the self assembling of phospholipids is dependent upon the packing parameter p. So the formulation of the two systems should be different. See for example:

and try to calculate your packing parameter.
If you already prepared it you can check the size of your system by Dynamic Light Scattering: usually micelles are smaller than 20 nm.
methods to 'create' micelles are different from creating liposomes. Micelles for surfactants able to form them are obtained by surpassing the critical micelle concentration. This is simply dissolution of sufficiently high amount of surfactant. These are thermodynamically stable solutions. Do not know what happens if peptides are conjugated on them.
