When performing Cas3/7 activity assay using luminescent substrate cleavage, Caspase-Glo® 3/7 Assay as an example, and according to the product protocol, the signal is proportional to the cell number. In any treatment or perturbation of the cells, a change of cell number is always expected. However, non of the references i came across, including the kit manual, mention about this . My assumption is that the signal needs to be normalized to the cell number i.e. cells need to be counted prior to the assay! any suggestions or tips regarding this?
