I'm currently collecting stool samples for Rotavirus ELISA assay. Please how do I preserve the stool samples until number of samples needed are complete and how long can I preserve these samples before analysis?
preservation of stool samples depend on the type of ELISA you are applying. In case of detecting antibodies in stool samples , you can make aliquots of stool sample nd put them in dee freezer at -20C for 1 to 2 weeks test them with ELISA. But if you are detecting antigen in stool sample by ELISA, it depend on the sensitivity of the targeted antigen. However, I advice you to apply internal quality control check that you will divide the samples into two aliquot (one fresh and the other one freezed at -20C) and test the fresh stool sample with ELISA and keep the result; then test the frezeed sample with the same test and compare the obtained results to validated the process.
Use glycol saline solution for the preservation of stool sample. The viability of the microbes within the sample can be maintained when preserved in the aforementioned solution.
Thank you all for your feedback. I'm trying to detect Rotavirus antigen and I have to wait until the number of samples required for 96-T ELISA assay is obtained. Should I aliquot using saline and then freeze at - 20°C? Mahmoud Hamdy Mustapha Umar Andreas Eisenreich Thanks in anticipation
