I want to see the nanoparticle that embedded in fiber(300nm diametar) and I do not know what type of grid do I have to use in order to have good contrast between the nanoparticle and fiber which are both natural polymer and how do I prepare sample
can I use a copper grid of TEM on the electrosppining collector and collect thin layer of fiber then get TEM image from that?
What you suggested works really well. Use a copper grid as target and electrospin onto it. However, the only thing that makes me concerned is how much densities of your fiber and nanoparticle (if both are polymer) are different from one another. In case they are very close, you might not see a recognizable contrast. Therefore, I suggest you to try SEM first if you have not yet, due to the large size of nanoparticles you are using I am sure if you have enough density difference between the fiber and the nanoparticle you'd be able to see both; even though you have to make sure you are using an enough low electron density in order not to melt the polymer.
