Hi, new member here... I am currently having difficulty dissolving HBTU in DMF to create a .2M solution. I also add NMM and oxymapure but those substances all seem to disolve except for the HBTU, most of which precipitates and lingers at the bottom of the storage vessel. Ive checked my math and tried mixing it in different ways but i still end up with the same result. Is there a special trick for getting it properly mixed or should i go back and check my math or instructions again?
firstly, why do you need to create the solution? if you want to activate the acid and couple the substance, you may not care the precipitate, which will disappear during the reaction process. secondly, you may change other condensation agent, such as HATU for amide, DCC for ester.
HBTU is a salt an may very well not be that soluble in your solution. It may react with your material anyway but you would have to add the appropriate amount of HBTU into your reaction rather than making a solution of it. Otherwise you could use other reagents, like HOBt and another carbodiimide (the combination would be equivalent to HBTU). There are many coupling reagents known and you might have to screen a few to select the one that works best for you.
