I need some advice to perform a drug test on breast cancer cells. I'd like to evaluate the cell's viability after treatment by MTS test. Do someone have some protocols to understand more.

many thanks for your advice. when I seed the cell in 96 wells plate, do I have to wait 24 hours to make the drug treatment or it is better to wait the double-time (DT) of the treated cells? thanks
Dear Nicolò Rensi It is ideal to wait for the double time of the cells. However, I do not see any wrong of doing it after 24 hours period if you are using an acceptable cell count number per well. This acce cell count number per well can be refer with the manufacturer protocol usually (In the manual for reagents). But please make sure to use the correct 96 well plate type. Esp you may need 96 plates that are treated which can adhere cells In to the wells. Otherwise cells can get out from the media during the media transfer. Yes Nicolò Rensi , you can calculate IC50 once you get the toxicity/viability data. I would recommend you to use either graph pad prism or origin software if you have one of them. Otherwise you can even do it simply in excel. Make sure your NTS assay comes with triplicates which is ideal.
