I know this somewhat of a stupid question but I wasn't able to figure out the answer after many trials and research. The brain slices are in an anti-freeze solution, so we use Tris-Buffered Saline solution to rinse (takes about 30-60 minutes, depending on how many times we need to rinse it) the slices. Then we use either Phosphate-buffered saline or distilled water to put the slices onto the slide (we use charged slides to make sure the slices stick). Later we let the slides dry on a heater. After that we start the defatting portion of cresyl violet staining where we put the slides in distilled water, 70-95-100 percentages of alcohol and xylene. However the slices come off even at the first step of dipping the slides into distilled water (and we're being gentle).
We also tried using floating brain slices but because the brain slices shrink and solidify, we cannot mount them properly. So we need a way to make the brain sliced stick onto slides. Do you have any suggestions on how we should proceed on?
