How do we know whether the white rot fungi cultured on agar medium produces spores or only mycelia? The type of white rot fungi that I'm dealing with is basidiomycota species and known also as wood decay fungi. Some peoples said that its only produces mycelia and no spores when we grow it on agar medium. I need a verification about this matter.
From the wood decaying fungi that we are cultivating in our lab, none produces spores.
Thank you very much for that answer Dr. Philipp. So for the cultivation in liquid broth, we will use mycelia disc cut with cork borer. I have another question, how much mycelia disc i need to use to growth it in 100 ml liquid broth? Currently i'm using 9mm mycelium disc.
I am not working with liquid cultures, but I would assume you make a trial and find out yourself what works best.
You can find out if a fungus produces spores by stressing it. We usually use Tap Water Agar, to induce sporulation in fungi. Preparation of Tap water agar is done by weighing 15g of agar powder and dissolving in 1 litre of tap water then autoclave.

Yes, after sterilization and pouring as done usually with other media, inoculate as usual, as done in standard media and incubate at 25 degrees.Then observe after 5-7 days. You will be required to make a slide then observe for spores under x10 and x40,light microscope.I hope you get some results.
