Hello! I am in need of help analyzing my results from Gel Electrophoresis. The first photo indicates that there was DNA extracted but did not go down. The second one shows that the DNA has been degraded. I want to confirm my observations. And if ever, may I ask why these results came to be? Thank you!

It seems some of your DNA extraction component is contaminated with DNase either your extraction solvent, MQ or RNase. So try to prepare every thing fresh properly autoclave the MQ and other solvents. To check your RNase contaminated with DNase you can use any Confirmed DNA and give it RNase treatment at 37 degree C for 30 min and load on AGE if it is okay you will not see any change in DNA band but if it is contaminated you may get denatured or no DNA band at all. The 1st electrophoregram shows that the DNA might be contaminated. U repeat the phenol extraction twice or thrice and perform the ethanol wash twice and airdry the DNA pellet properly.
For the second electrophoregram, u have analysed it accurately, since the DNA seemed to have degraded. To avoid this type of thing, u autoclave the plasticwares you are using and if possible use freshly prepared solutions. If it is annoying to prepare the solutions, then you can heat treat the materials like phenol, rnase, water, etc. for 10-15 mins at 65C and let it cool down to room temperature, before you start the extraction

One thing more please check your AGE buffer components too there is some problem in that too because your ladder is also not clear and run abruptly.
