I have a problem with chromosome analysis, as they appear unclear. The normal chromosomes are not visible in the metaphase stage. I have been conducting this analysis for a long time, and it used to yield good results. Suddenly, all samples do not show the metaphase chromosomes. No solutions or procedures have changed. I hope to receive assistance from those who have expertise in this matter. Attached are images of the slides that have issues.

Please check quality of PHA and colchicine. It is cultured for 72 hrs or 48 hrs.
We culture it for 48 hrs, human lymphocytes (whole blood), and the colchicine treatment is for 24 hrs at very low concentration (AERB protocol) and get almost 1000 metaphases in a single slide.
Can I use colcemide instead of colchicine in this protocol (48h) or must use colchicine?
