It could be between lines as well. The lines and images are already registered. If I just do a subtraction there are a lot of differences due to noise. It seems to me that the way to do this would involve not only looking for which pixels have the largest differences but looking at the neighborhood of the pixels. The larger the neighborhood that has a large difference the more significant the difference.
