first remove mucilage from plat material using organic solvent.Then extract 2g of plant material with 5ml distilled water.Now to 2ml of this extract add 5ml 10% cold TCA to precipitate protein.Take the precipitate and digest it with about 2ml conc.sulphuric acid at low temp.(25-30*C)till it turns apple green.Now make the volume 5ml by adding distilled water.From protein can be estimated following standard method of Lowry et al(1951)or any other method

The extraction method here is very effective for tricky tissues.
Leaf apex of Portulaca oleracea is obtuse and for P. quadrifida it is acute. Color of petals in P. oleracea is yellow and for P. quadrifida it is pink. Moreover, capsule structure can also differentiate these species. I think these characters are sufficient to state difference between these plants.
