In my project I have biochemical estimations in peripheral blood lymphocytes. I isolated lymphocytes using Histopaque 1077 and the buffy coat was isolated by pipette. How do I go about biochemical estimations in lymphocytes?
Hi Mandiner , generally lymphocytes were analyzed for ability to grow in culture; surface and cytoplasmic IgM; functional activity in blastogenesis, cytoxicity, and suppressor assays; and biochemical activities for alkaline phosphatase activity, adenosine deaminase activity, purine nucleoside phosphorylase, adenine phosphoribosyl transferase, hypoxanthine phosphoribosyltransferase
AMP deaminase. So some of biochemical activities carried on cell suspension and other need growing culture.
The cells after isolation of lymphocytes I am dispersing the lymphocytes in hypotonic solution like water and incubating it for half an hour at 37 degree Celsius after which I centrifuge and the supernatant will be used to estimate GSH , lipid peroxidation , nitric oxide, catalase SOD Can i follow the above mentioned method for estimation.
