I used sterile distilled water to dissolves my extracts to get a concentration of 320 mg/ml. However, my aqeuous extracts do not dissolves completely in the distilled water. And when I want to filter the extracts for sterilization purpose (with 0.2 micron syringe filter), it became to difficult as the extracts is too concentrated, with the undissolved extracts. Then I proceed with centrifugation to separate the solution and the undissolved extracts. Did my final concentration in the filtered solution changes after centrifugation? And how can I increase my extracts solubility, so that I manage to get a homogenize solution, and easy to filter?
I do not know the nature of your extract, but anyway 320 mg/ml is a lot, It is not possible to dissolve such amount even if the extract is hydrophylic. Of course, the concentration will be lower after centrifugation or after filtering. To increase solubility, you can add some salt (for instance phosphate saline buffer) or some detergent to the solution, but anyway I do not think that you can solve 320 mg/mg. Good luck
I agree with Francisco Solano mainly and can add: if you want to dissolve hydrophylic and hydrophobic components both, you have to use water solution of ethanol, for example. If you want to use your dissolved extracts in living organism you can vaporize ethanol before use. Good luck
You can test whether the problem is just that the solubility of the dry substance is insufficient in water by seeing whether the pellet from centrifugation can be dissolved in more water. If not, your aqueous extract contains something that is no longer soluble in water. It may be soluble in water if you adjust the pH up or down, however. It's solubility may be increased by increasing the temperature. But as Francisco Solano pointed out, 320 mg/ml is a very high concentration (For sucrose, that would be nearly 1 M.)
If you evaporated your water extracts to dryness I think I would use an ethanol/water mixture for the resolution. But without knowing the specific problem.. I can be of little help.
You could try DMSO. You could mix solvents, as water and ethanol, water and glycol (butylene glycol, for example), water and glycerin, Sometimes, the order of addition of the solvents can interfere in the solubility of the compounds, so,you can try to add all the solvent (previous mixed) to the sample at the same time. You can also try to add first the one, mix everything , then add the other, etc. Good look.
