i used disc diffusion for screening of antimicrobial activity of a plant extract, and I used concentration range 100 mg/ml to 20 mg/ml. For each concentration, no zone of inhibition were observed except for positive control (commercial antibiotic disc). However, when I do Minimum inhibitory concentration test (MIC), I get positive MIC result, at concentration within the range as in disc diffusion test. Why can this occur? Is there any articles/journals that can help me in explaining this issue? thanks in advanced!
It was the same problem for me at my first experiences.
After many tests it seems that:
May be your extract is hydrophobic and can't diffuse in cultural media.
Or may be your extract easily evaporates, so any apparent inhibition zone.
Some extracts can not be used for disc diffusion method, and there is no real relation between the two techniques as I find in my work.

I find this in my practice but I don't know a paper on it, but I have never mention it in my articles only in my master thesis.

Try to find other researchers on research gate who work on antimicrobial plant extracts and ask them this question.
