Currently planning an experiment on the expression of C-Myc in individuals struggling from POEM Syndrome. Just wondering how I calculate and determine the primer volume to use in qPCR
The qPCR mix will come with suggested primer concentrations. You'll have to dilute them to a working concentration (typically 5-10 micromolar) so that you are pipetting volumes that are easy to accurately measure to create the final concentration in your reactions. When in doubt, use the c1v1=c2v2 formula. Good luck!
When you received your primers, you found a paper along with primers mentioning how many µL of water you have to put on each tube. Normally at the end you have 100umol and I used my primers at 10umol for the PCR.
I used to make 1/10 dilution of my primers before using them.
