Hi, I am working on antioxidant property of medicinal plants, extract prepared in hexane and chloroform (non polar solvents). While working with these extracts, i am facing a problem as all the reagents of different tests (phenol, flavonoids, FRAP, DPPH, ABTS, etc) are prepared in water or methanol (polar solvent), and these non-mixable with plant extracts (hexane and chloroform extracts). so when i added extract and reagents together two separate layer formed and absorbance of this mixture also misleads. So Can anyone suggest me any solution to overcome this problem?

Plant extracts a complicated composition, but not all react in such a way that they form two layers including a single emulsion. Maybe try for the help of distributors. Water is a good solvent in terms of permeability if the substance.

Yes these all antioxidant tests are performed usually in methanol and in my experience i saw that hexane or chloroform extracts are not micible in methanol but i solved this problem by continuous stirring on aplate for several time until become homogenuous mixture

How and what amount should i use of Tween 80? does it affect the property of plant extracts?
