I need to add Gentamicin to cell culture media at varying concentrations in order to study toxicity. I have a 1g bottle of powdered Gentamicin sulfate (from Sigma). In the description, it said that this product contains three types of gentamicin (C1, C1a and C2 with molecular weight is 477.6, 449.5 and 463.6 respectively). Why to use the molarity? In such case it is preferable to use the potency (μg/mg) to prepare different concentrations in (μg/mL).
This drug has been used in my specific cell line in the literature, and in all cases, the dosing is reported in molarity. To be consistent with the concentrations reported in the literature, I would like to use the same dosing.
If you do not find any detailled information concerning the various types of gentamicin I would suggest to calculate the molar concentrations by declared amount of gentamicin base. Make sure that the ratio of the three components is documented.
Other researchers must have done the same considerations as you do now as you use a standard. Is it a option to contact authors who have worked with this standard?
