I am starting with a drug assay and I need the count of dead and living cells and also the total count of cells in my wells. How can I do that with flow cytometry?
Generally flux citometers have a function to count cells or determine the concentration of the suspension. You should look in the manual for it.
Also, you can make a region in the population of interest (based in FSC vs SSC parameters, for example) and check the number of events.
If you have a way to determine the flux speed and the total running time you can get the concentration too (and indirectly the number of cells in the suspension)
You could stain with Propidium Iodide and then do flow cytometry - the PI will stain the dead cells but not the live ones and then you can count the 2 populations (just remember to not fix the cells first)
LIVE/DEAD® Fixable Dead Cell Stain Kit is useful, dead cells and live cells can be distinguished distinctly by flow cytometry. While dead cells will be stained but the live ones will not, nevertheless, the kit may be a bit expensive.
