Hi, I have done some gel-filtration chromatography and now that I want to plot my results, I am not sure if it should be UV absorbtion per time (min) or UV absorption per volume? Does anybody have an idea about that? are they really different scientifically?

Can you elaborate please on the experiment setup? did you measure UV online or off line? if offline, were you measuring fractions? uniformly collected fractions?

Thanks for your reply David! I measured the UV offline With the program designed for the system (which I am not totally familiar With). And I collected all the fractions. So I have the data on fractions and volume as well. But what I am wondering is that which one I have to use when plotting my daya (on the X axis). Since I can see both types of results in papers. :)

There is no right/wrong answer here. I would use volume, as your not measuring instantaneous concentration but rather an average over time span. but it's really up to what you feel more comfortable with.
