I am having challenges understanding a certain part of the protocol which is being used to test urease activity with the aid of the conductivity meter. I would like you to assist me with this problem.
If conductivity reading (mS) was taken every minute for a duration of 5 min and was used to generate an average change in conductivity (mS/min) and is used to determined urea activity which is mM urea hydrolysed.min-1 = Conductivity(mS/min) x 11.11 at 25˚C And specific urease activity was calculated by dividing urea activity by optical density @ 600nM , what OD value should be used in the calculation to determine specific urease activity?.. Should OD be taken after conductivity reading is done at 5mins or a OD should be measured every min and an average OD value was be used?
