I have glutathione reductase enzyme from Santa Cruz. the data sheets specify following information. 120units/mg and conc is 5mg/ml. vial of enzyme only 120units/mg in ammonium sulphate suspension and donot provide info about amount given in vial,
I want to use 0.05ml of glutathione reductase having 1unit/ ml for antioxidant assay.
how will i make this?
For preparing 1 unit/ml, you take 5 µl (microlitres) of stock solution (5mg/ml) make up the volume up to 3 ml with distilled water or solvent ( 5 µl stock solution + 2995 µl of solvent or distilled water). By this you will get working solution of 1 unit/ml.

(i) 120 units/mg
(ii) 5 mg/ml
that means you have 600 units/ml
To prepare solution with 1 unit/ml, you need to dilute 600 times [that means you can take 1 ml of 600 units/ml and add 599 ml of solvent (preferably a buffer or distilled water)].
You can take 10 µl of 600 units/ml solution and add 5990 µl of distilled water or buffer or as suggested by Mr. Ratendra Kimar you can take 5 µl of 600 units/ml solution (as provided by the company) and add 2995 µl of distilled water or buffer.
If vial has only 120 units, then the vial must be containg only 200 µl of suspension. We prefer injecting 800 µl chilled buffer or distilled water into the vial and vortex. In such a case, we will have 120 units/ml GR solution (you can dilute this to get 1 unit/ml).
