One of the main issues in better acclimatization of in-vitro woody plants is "stem thickness". How can we get the ticker stems during multiplication stage?

Not only the light intensity but also the the quality/source of light influence the growing shoot..for example:BLUE light strongly inhibited the elongation. Incandescent light promoted both growth of stem diameter and elongation of leaf petioles (From:Reference+my own experience)

Sreenivasan Ettammal In our experience, blue light give twice shoot thickness compare to red light. May be a combination of blue+red light be a better choice for getting thicker shoots. What's your idea?
Carbohydrates source in combination with light intensity might be helpful to increase girthness of in vitro shoot instead of GA3.

Contrary to some comments, GA3 and BA do not have a positive effect on stem thickness. At first glance, the application or increase of BA level may improve the thickness of the shoot, but these stems are hyperhydric and have difficulty in acclimatization because of water loss. Substituting TDZ or adding adenine sulfate to the proliferation medium achieved better results. Based on personal experience on three plants (R. damascena, Vaccinium and Cornus) three important factors increase the thickness of the in vitro shoots. Increasing the concentration of calcium in the culture medium, specifically through the adding calcium gluconate, raising the concentration of carbohydrates (especially a combination of glucose and sucrose) and reducing the level of ammonium in the culture medium and substituting it with nitrate (calcium nitrate is very effective in this regard).

Dear Kourosh Vahdati To my opinion, stem thickness is a heritable trait with a large extent of genotype x environment (composition of media, culture condition, the duration and quality of light energy, size of population, and the like) interaction. Optimizing, controlling and validating these variables are expected to provide desired results.

Yes I do agree with Hamid. Based on my experience TDZ hormone induces greater stem thickness. But you may try also other types of hormones which is suitable for your plant.

you may use silver nitrate or carbon nano tubes, a secondary result is the shoot thickness.

To increase stem thickness of woody plants in tissue culture condition you have to use indole acetic acid spray .
In our experience, after optimizing the growth regulator concentrations, the application of moderate seismic stress under in vitro conditions improved the thickness of stems in some woody plants.
