How can we decide the concentration of the buffer in the mobile phase? And the effect of concentration of buffer on retention?

Dear shrikant,

Concentration of buffer plays very key role in determining your chroamatographic requirement. High concentration reduces your column life also. Even though some times it may require depends on our molecule,in general 10mM to 20mM is most preferable to achieve good chroamatographic conditions.
