I want to calculate the amount of Proteins in unknown sample with A = 0.059 at 595 nm by Bradford method. I use BSA for standard curve and plot graph on excel as polynomial curve having 3 order points on trend line with equation and regression as under.
y = 4E-07x3 - 0.0001x2 + 0.0087x - 0.0051 R² = 0.9945

In your case solving for x will give protein concentration ( y = ax2 + bx + c ).
Logic is same like solving polynomial in the high school.
Here is my alternative strategy :
I plot protein concentration on y-axis ( dependent variable) and absorbance on the x-axis (independent variable). In this way concentration is represented by y instead of x, and it is very straight forward to calculate
A second order polynom is probably sufficient for your purpose, you don't need third order.
