I have to do a research proposal to follow a recent research.
I chose a paper where they found that a High copy number of a gene is correlated with an obese phenotype in humans.
The next step is to see whether the expression of that gene specifically in the adipose tissue is correlated to obesity.
I was thinking to use a mice model in which I insert x copy number of the gene and then, knockout it in the adipose tissue with a cre lox method to see is it contributes singnificantly in the obese phenotype.
I don’t know if that is too complicated or if there is an easier way to go.

Generally, the approach of using a mouse system is possible. However, did you really put enough thought into it? You are talking of the amylase gene, right? What makes you think the effect on adipositas is due to overexpression in adipocytes? From knowing what amylase is doing, I find that rather unlikely, actually. After all, it's a digestive enzyme. Is it really expressed in adipocytes, no matter what the copy number is?
As Bodo said, a mouse system is always a lot of work. Get a good hypothesis first. Always have some experimental evidence from other systems first. Also note that there seems to be a lot of work on mouse amylase gene, including transgenic mice, coming from way back when (80ies)......;-)
