I extracted star apple stem bark with Ethanol by maceration. recently, I've checked phytochemical compounds with the present of tannin, alkaloid, terpenoid, saponin, phenolics, cardiac glycoside, ascorbic acid. I'd like to find a mobile phase to separate the compounds on TLC plate. I try with hexane:chloroform; chloroform: dicloromethan, toluen: EtAc; but they show 1 or 2 band only. What should I do? Thanks
Too many compounds with different polarities for just one TLC. May be you have to try different conditions for different compounds. You have used the most usual solvents for apolar compounds. Try mixtures. See for instance

Ethanol is high polar solvent.....so that you have to try with low polar solvents combinations like chloroform:ethylacetate/methanol, then you change ratio and standardize ..............
Ethanol is a very polar solvent and will extract polar component. You should try to test whether your crude contains saponins is polar hydrocy compounds. Try CHCl3-MeOH-H2O(64:50:10), n-BuOH-gl. CH3COOH
Thanks all. I ran TLC with your recommendable conditions, still observed one band. I have set different gradient with several solvents and combinations. nothing change.
One thing, when I evaporate my extract, there is an very sticky residue formed. Is it latex? I think. because my plant (star apple) has rich of latex.
