I linked amine functional to Polar compound that contains acid group. I need to purify the compound but it is not soluble in organic solvents and I tried TLC (10,20, 50% Ethylaceate/hexane, 5% methanol/DCM, and chloroform/methanol. but I am not able to get clear spot and while running NMR and Mass so many impurities. Can anyone suggest solvent system for polar compounds?

Hi..u can start with chloroform with a few drops of methanol as mobile phase, then increase the polarity of solvent system. Also can try mixture of acetone and ethyl acetate.Good luck
You may try with addition of little trimethylamine into your eluent system that helps to elute the acid functional compound.
Why don't you try with reverse phase TLC. run with Methanol or methanol +0.1% water. RP silica is also available. you may try.
50 water:50 Methanol, usually reverse phase silica(c18) is best for isolation of polar molecules

First you have to try with chloroform and methanol(9.5:0.5), then gradually increase the polarity of your solvent system by adding methanol.
Combination of Toluene: Acetic acid: formic acid (Vary ratio) also actually better for separating compounds using TLC
Butanol:Acetic acid:Water (7:1:2) works well for many polar compounds
Atolani Olubunmi (Ph.D. Chemistry) may I know which type of of TLC plate can be used? i already tried TLC F254nm, however it does not separated well
