In the recipe of a tissue culture media, I need to use 27.85 mg/l ferrous sulfate heptahydrate and 37.25 mg/l disodium EDTA dihydrate. My lab has FeEDTA salt stock but not the above said two salts separately. How can I use the FeEDTA salt instead of the above salts and in what ratio to those two separate salts, I should use FeEDTA?
Ther purpose of FeEDTA is to furnish Fe, avoiding its precipitation. So youjust calculate the amount of Fe in moles , here 27.85/278.02 = 0.1 mM so you just need 0.1 mMol of your FeEDTA solution for 1 l tissue culture media.
you can use 36.70 mg/l FeEDTA
EDTA is frequently used to prevent FeSo4 from chelation. easier is to use FeAEDTA instead.
for Ms culture medium 27.85 mg/l ferrous sulfate heptahydrate and 37.25 mg/l disodium EDTA dihydrate is recommended while in B5 culture medium 36.70 mg/l FeEDTA is recommanded. this form may be absorbed faster.
