TEM can show layer of ur sample and AFM can help u to measure thickness of each layer.

but exist one form to determine the number of layer by Raman spectra ? some times many people report the Increase the FWHM increase and another people report blue and red shift and its assosiated with number of layers
You may get an idea about the number of layer of Graphene with the help of I2D/IG ratio in Raman spectroscopy. Like the image is showing.
