I want to see level of glucose and quantify the same in C. elegans using LC-MS/MS . Please suggest me the extraction procedure as well as LC-MS/MS conditioning for the same.
The method of choice is mass spectrometry as you well-know.
However, the quantification should be carried out within the framework of our own-authored (to me and my co-author's theory according to the authorship shown below) innovative stochastic dynamic concept and model formulas for quantification of the measurable variable 'intensity', which we have developed and published more recently. Only this approach, currently, worldwide provides exact quantification of analytes at very low concentration, thus exploiting the superior method performances. All other available theories, including those used to the current Directives for quantification by mass spectrometry operate at lower capability of the method (at about 98 %.) (Please, see more detail on the introductory section of refernce [2].)
Our method has been tested, so far, of analytes in mixture at pg.(mL)-1 concentrations, showing correlation between theory and experiment /r/ = 0.99997-1.

In particular, our theopry has been tested on mixtures of carbohydrates in reference [3], showing reproducibility of the quantitative outcomes /r/ = 1 in chemometric terms within replicates of 6 samples; or, our resuilts are 100 % reproducible.

The method is valiudated using chromatography, as well.
The calculation steps according to our theory and formulas are detailed in reference [3].
Also, our theory has predictive capability, as well. In other words, via its application there can be determined uknown analytes only looking at its mass spectrum.
Our method has been tested successfully on different ionization methods, as well. It is applicable to both solid-state (MALDI) and solution (other MS methods) condensed phases.
