I see that some pCAMBIA plant expression vectors feature histidine-tags. I am having a difficult time finding examples of purification of tagged proteins from plant tissue.
I have read examples of protein secretion from root cultures, however I cannot find examples of purification of His-tagged protein from plant tissue.
Are there known issues with grinding up an entire Arabidopsis plant, clarifying by dialysis or membrane filtration, and passing through an Ni-NTA column?
Theoretically, once you dissolve the cell wall and any other connections between the cells, it should be a normal cell for the most part. Perhaps you should look for cell wall removal protocols and then use a standard homogenizer / probe sonicator protocol.
Alternatively, I recall that NaOH can be used to dissolve cell wall in fungi. Since histag proteins have been overexpressed in yeast before, there may be relevant literature that could help.
