In my country, Bangladesh, V8 juice is rare on market. I intend to prepare V8 juice manually however all 8 vegetables are also hard to find in here. So, is there any chance to prepare a suitable media for Phytophthora isolation using locally available vegetable juice such as spinach, carrot, cabbage, tomato etc.
I am referring V8 agar media because as per my knowledge V8 is the most suitable growth media for Phytophthora. I am in a fix. I would really appreciate experienced help.

Md. Abdur Rahman Tomato juice takes 87% of basic V8. There are many variations in V8 composition, but commercial V8 often has chemical conservants. You can add 13% of other juices (spinach, carrot, lettice etc.) and test the compositions.

I agree that probably many types of (tomato based) vegetable juices will work as well. I have used so many, different brands, different compositions. Tomato juice alone will most likely work as well, amended with CaCo3. You can try small batches with existing isolates.

you can try this method
80 gr Tomato
20 g Carrot
12 g Beetroot (Red)
4 g Lettuce
2 g Celery
2 g Parsley
2 g Cress
2 g Spinach
After boiling, we filter it with the help of cheesecloth and make it up to 1 lt with water. It will be autoclaved by adding 3.0 g of CaCO3 and 15.0 g of Agar.
I use it instead of 20% V8 juice agar.
In one company where I worked, all media production for Phytophthora inoculation was made using industrialized tomato juice, here we called "tomato passata".
In another lab, we changed the V8 media for CA (Carrot-agar) but made the carrot juice, double-filtered in "voal" fabric, and Phytophthora grow and sporulation very well.
Hi since many years I'm using rye medim for P. infestans growth. It is much better to grow for P. infestnas sporangia isolation compared to V8. So you can try that.
