Hi all, I purchased 25 mg of GSK126 (HY-13470) for combination treatment in BALB/c Nude mice (A549 xenograft). Literature shows various dosages ranging from 50 mg/kg to 300 mg/kg for different xenografts. However, I also noticed that 50 mg/kg is considered as "exceeding solubility."

According to my calculation, the required working solution concentration is 31.1 mg/mL, which is about three times higher than the 10 mg/mL solubility reported from the reagent supplier.

I am confused about what dosage I should use and how to prepare the solution:

  • Can I use beta-cyclodextrin hydrate to dissolve this drug? If yes, what concentration should I prepare?
  • Is filtration necessary after suspension preparation?

Any suggestions would be greatly appreciated!
