I need a method for counting the stomata and epidermal cell number of a herbarium leaf with a very dense trichomes:

You can give it a try with agarose imprints (that, besides, shouldn't damage the tissue).
The method comprises applying a layer of clear nail polish to a surface of about 2 cm2 leaf. Once dry the varnish became a colorless tape stuck above, then removed and placed on a microscope slide to be observed microscopically.
Considering the large amount of trichomes that completely cover the stomata was applied on the underside of the sheet tape repeatedly until the complete removal of these structures.
Try to hydrate the small part of leaf with glycerol and then try to peel the epidermis
after removing the trichomes by a sharp razor you could use of KOH AND H2o2 for peeling the epiderm
Hydrating by boiling tissues with water and gliceryn, then under a microscope, remove trichomes with a razor blade carefully. To separate epidermis you can use chromic acid and nitric acid or chlorine bleach and heat at 60˚C depending on the condition of the material
