I have developed a lateral flow assay using the same monoclonal antibody for both capture and detection. The assay consists of three lines: one control line and two test lines (T1 and T2). While I have successfully optimized the T1 test line, I am facing challenges with T2.

Despite testing various antibody concentrations for the test lines, the T2 line develops even at lower biomarker concentrations. If I dispense a lower concentration of antibody for T2, the line dosent appear even at very high biomarker concentrations. However, if I use a higher concentration, the T2 line develops even at very low biomarker levels.

How can I optimize the T2 test line concentration so that it appears only when the biomarker concentration in the sample exceeds a specific threshold?
