I need to know the surface roughness of fresh tumor tissue. Has anybody ever done that? Is it just similar to measuring the surface roughness of a glass or metal surface?
Atomic force microscopy (AFM) measurements and scanning tunneling microscopy (STM) technique are used to determine the surface roughness. I recommend a few articles:

In addition to Dr Houssem Rezgui suggestions, you may see out https://www.researchgate.net/deref/http%3A%2F%2Fwww.mitutoyo.com%2Fpdf%2F1984_Surf_Roughness_PG.pdf
Go for Atomic force Microscopy it give tropoghaphy of the tissues. Even You use the Advance AFM instrument they produces three dimensional measurements too.
