You need antibodies against IFNg to measure it using ELISA. Sandwich ELISA is more reliable to quantify a protein in supernatant. If you use a sandwich ELISA, you need 2 antibodies to sandwich the target protein between antibodies.
Thanks a lot Suhee but where can I get this from? Actually nobody has developed mabs against IFNg as well as not against IL10 which is possibly much better for ELISA compared to IFNg. All I can find is the detection of the genes on RNA level. This is not sufficient to me, very theoretiucally only. Therefore my request for help. :-)
I never known aout the kits. It is very thankful that there are ELISA kits to detect fish cytokines. However I wonder how they developed the antibodies against "fish" cytokines and how guaranty their specificities.
I think you should ask to company about guaranty. Because I used only once and I don't have any chance to comparison repeadibility and accuracy. But There are several publications where these kits are used.
Thanks to all of you. I even was told that will not be possible because the suorce of IFNg is the macrophage not simple cells. Therefore it is no sense to do so. What can be done is IF alpha or beta which is expressed at least on mRNA level by different cells also by cultured cells.
