HI, everybody. I have a problem like Tatiana. My drug is fat soluble, but my nanocapsules are emulsion from oil and water soluble material. I would like to measure encapsulation efficiency by spectroscopy method because we don't have HPLC in the lab. My drug has many peaks (4 peaks) in absorbance graph at difference landa. Can I request to help me please?
Dear Shahmoradi, what I understand from your question is that your have a lipophilic drug which is dissolved in oil to form oil-in-water emulsion. If this is the case, either you can measure amount of drug in oil by dissolving oil droplet (separated by centrifugation) in a nonpolar solvent or by measuring amount of un-entrapped drug in aqueous supernatant. If you get 4 peaks in UV-Vis spectrum, it is normal. You should select the peak with highest intensity i.e. lambda max. If you feel that peaks are similar, you can proceed to prepare standard/calibration curve With all peaks and see which is most linear because some wavelengths do not give straight line at very high or very low concentration.

I think the given answer by Dr. Mubashar Rehman concerns the encapsulation capacity; on the other hand if it is meant the encapsulation efficiency this must be measured periodically at a prolonged time; of course by the same mentioned techniques. Hi. I would like to measure encapsulation efficiency .
As I understood the main issue is the complex medium!
So, I suggest using Dialysis tube or bag. Through the molecular weight of the drug choose a dialysis tube with a proper molecular weight cut off (MWCO). Undesired molecules will stay inside the tube and the drug will release from the bag to the exterior medium.
So, Pour the cocktail in the tube and at different time intervals measure the drug concentration in the released medium using UV/Vis spectrophotometer (if there is any absorbance!).
Please note that you should put neat drug solution inside the bag as well, to see how much of drug is entrapped in the tube!

drug efficiency must be evaluated as a function of time, with membrane dialysis or accelerated by centrifuge. Encapsulation efficiency will obtained if the system is experienced for a prolonged time. Regards
