I have made polymer films on glass substrate through spin coating and then tested them against E.coli using JIS Z2801 protocol. No colonies observed in any dilution. How can i make sure that bacteria are growing on polymer surface? What should i use as control as i have observed E.coli did not grow on glass?

Dear Shazia Nawaz, a valid query and interesting also because of its two aspects:
1. As a tool of preventive measure to understand about surface contamination ;
2. It could be a sensor also (if you are able to speak about the microbial growth or even its presence)
The protocol you discussed is well standard for as per International norms, in which you observed No colonies.
It has been observed by some of the researchers to utilize Basic pH indicators as control tools to observe activities of microorganisms. So you may use this also. Ecoli usually releases hexanal in their metabolic activity hence indicator for the Hexanal may give you some hint about presence or absence (but nothing more than that).
However, as a worker of Lipid polymer, it is my keen observation that few of the polymer itself has the inherent capability against some specific microorganisms.
eg: Chitosan (against aspergillus,), Shellac (Ecoli and fungi). Please check your polymer for the same using a generic method.

To verify this, the plates must be compared before and after development of bacteria on the surface of the polymer, by UV-visible spectroscopy. It is just enough to expose the polymer films deposited on the glass substrates, as samples to be analyzed in the UV-visible spectrophotometer (cuts the glass slats with dimensions of 0.1 cm x 3 cm, to be able to introduce them into the sample holder and to be able to put them in a standing position facing the light beam). The other way to verify this is to observe the surfaces of the polymers under an optical microscope.
