I am not familiar with growing MLV, but your titres already seem quite high to me. Have you ever considered concentrating the virus by ultracentrifugation through a sucrose cushion? While I know that this does not work for HIV-1 due to shedding of the envelope protein, it should work fine for MLV.
There was a good correlation between the increased viral production and the titer measured after transduction. This improvement is of general interest, since a major goal for gene therapy is the production of high titer for increased transduction efficiency. The effect of different activators such as the glucocorticoids responsive element (GRE) are known to enhance expression of Moloney murine leukemia virus (MoMLV). The using of dexamethasone, and sodium butyrate (Na-But) alone or in combination may be increase titre of MLV. Pages JC et al (1995) found that 5 mM Na-But, or 5 mM Na-But plus 1 microM dexamethasone significantly enhanced viral production. These compounds could induce a 10-fold increase in viral production.
