1- aerobic growth (this will exclude the anaerobic actinomyces e.g. A. bovis and a. israellii) with characteristic actinomycetes morphologies i.e. rough chalky adherent colonies with soil odour.
2- Ziehl Neelsen stain: you observed non acid fast branching filaments (this exclude Nocardia and mycobacteria and other mycolic acid containing actinomycetes with partial acid fastness)
3- I subject strain to growth at 50C (psositve growth will exclude Thermoactinomycetes and Saccharmon/ polyspora species)
4- You left with Streptomyces, Actinomadura, Pseudonocardia, Amycolptes, Nocardiopsis-
These can be differentiated from one another but more tests are needed.
ALaternatively, and this is I found a short firm cut
B) Genotypically:
perform sequencing of the entire 16S rRNA gen and do phylogentic analysis
