I am looking for some biochemical tests that are different between these genera.
My procedure start by observing
A) Phenotypically:
1- aerobic growth (this will exclude the anaerobic actinomyces e.g. A. bovis and a. israellii) with characteristic actinomycetes morphologies i.e. rough chalky adherent colonies with soil odour.
2- Ziehl Neelsen stain: you observed non acid fast branching filaments (this exclude Nocardia and mycobacteria and other mycolic acid containing actinomycetes with partial acid fastness)
3- I subject strain to growth at 50C (psositve growth will exclude Thermoactinomycetes and Saccharmon/ polyspora species)
4- You left with Streptomyces, Actinomadura, Pseudonocardia, Amycolptes, Nocardiopsis-
These can be differentiated from one another but more tests are needed.
ALaternatively, and this is I found a short firm cut
B) Genotypically:
perform sequencing of the entire 16S rRNA gen and do phylogentic analysis
You will easily see where your unknown where it clusters or stands as new taxon..

Dear Farzaneh,
Streptomycetes can be separated from these genera (Actinomyces and Nocardia) by differences on the isomer of diaminopimelic acid (A2pm).
Streptomyces contain major amounts of LL-A2pm whereas the other genera produce meso-A2pm.
A simple method to determine the isomer of A2pm was described by Staneck and Roberts (1974; Appl Microbiol 28:226-231).

this is so simple if you find person who teach you knowing these genus by your eyes.
by the way, you can use plant bacterial books such Schadd 2001 and klement 1996
in these book you see very simple methods to identify this bacteria.
And I have many of these bacteria isolates in my lab.
if you have them use and and if you don't have any of them say, I can upload them to you.
