The tlc plate show pink, sky blue and purple color under uv light. is there any standard through which i can read that this color show this compound? and also there are some compounds which show their presence very low under uv. how can i visualize them?
Maybe have the sample analyzed using a properly developed LC-DAD-MS method (or whatever techniques are appropriate... GC, FTIR, etc)? Contact someone local who is experienced in sample identification and analysis for assistance.
What is the source of the sample? If the sample is the product of a reaction, run the plate with starting compounds, the mixture, and if possible, a known sample of the product. So long as the retention factor of a compound is different in the reaction mixture, you can state that you made something different, and also identify the spots corresponding to the reactants if one of them is in excess, or the the reaction is incomplete. This will not guarantee the spot corresponds to the compound you want. The TLC can be used to develop a column chromatography method to purify the compound for further analysis. There are also TLC- mass spectrometer interfaces commercially available.
If the sample is a plant extract or another unknown, the best you can do is exclude some possibilities on the basis of retention factor, and the TLC can be use to develop a column chromatography method.
If you are working on natural products, you have to 1st read about the nature of the compounds in the genus. For example flavones appear dark purple under UV, phenolic acids and coumarin blue, and so on
