i saw some black dots in my C2C12 cell culture. These thing are uneven shape and size. No active movement, is it contamination???Here I attached some pictures. Please give me some feedback and advices to handle the problem.
I see no evidence of contamination. Your images show phase dark, spread, cells, birefringent rounded up dividing cells, an aggregate of cells and two dark smudges out of the plane of focus. The dark smudges maybe on the outside of your culture dish or on the microscope optics. Am I missing the objects of concern? Arrows on the images would help.
Many thanks Ellen A G Chernoff, I am asking about encircled structure on plate, is that contamination?? because previously I saw 2-3 round bodies in culture plate but after few passages it increases approximately 3times...suggest me either can i use these cells or not?/
i understand. C2C12s can form aggregates. I would suggest that you detach the aggregates with a needle And discard before subculturing so you do not select for aggregation And disperse the cells well. I think that would be OK.
Hello, C2C12 is an immortalized cell line therefore the cells are only one step from becoming transformed i.e. uncontrolled growth. Indeed as you culture them it is more difficult to keep them myogenic than to have them derived towards a transformed phenotype. So as suggested by Ellen Chernoff: better get rid of this dense cluster of growing cells (if you can) or start another culture of C2C12 and not let them become confluent.
