I am trying to isolate adipocytes as well as SVF. however I find it very difficult to separate adipocytes from the oil layer. in addition it is hard to get a pellet of SVF. Can anybody help?
You should first very carefully remove the oil layer with a syringe. The adipocytes should be floating immediately below the oil layer; isolate the adipocytes with a large bore (e.g., 1000 mL and cut) pipette tip. The SVF can be isolated by a 500 rpm spin for 1-3 minutes in a 15 mL tube.
Thank you Jordan for your answer, I succeeded in removing oil layer, although it is exhaustive. but I am facing a problem that I do not get a pellet with SVF. what do you think is the reason for this and how to fix it?
