I wish to perform FACS analysis of leukocyte populations in tissue-derived single-cell supsensions. Is there any way one can get rid of blood leukocytes that interfere in the analysis? I want to use human adipose tissue, so perfusion of the main organ vessel with saline (like in animals) is not an option. Anything else to propose? Please also take into account that tissue amount is limited (~500 mg).
I haven't tried it, but maybe you can deplete them by MACS with specific antibody (CD45)?
Thanks for the answer. This would not work. I need to keep tissue leukocytes in my single-cell suspension. By using CD-45 depletion I would remove all leukocytes (from both tissue and blood).

Unfortunately our tissue is too small to perform such a perfusion procedure, as there is no "main supply artery". It is just a small piece of adipose tissue, containing small capillaries... Regarding any specific markers, again I am afraid this is not possible, since we just want to analyse infiltrating leukocytes, which actually have the same markers as the circulating cells they derive from. Maybe the extensive washing would indeed partly solve the problem.
