Hi,

 I have to do a bacterial conjugation experiment for the first time. My donor strains are fosfomycin resistant isolates of klebsiella pneumoniae and the acceptor is E. coli J53. However I have no colonies on the conjugation plates after the experement. I hope there is just a problem in my procedure somewhere. Here it is:

  • Inoculate donor and acceptor strains in 5 mL LB and grow overnight.
  • Add 0.5 ml the overnight culture into 4.5 mL LB broth. Incubate at 37°C for 4 h.
  • Add 0.5ml the 4 h culture of the donor and 0.5 ml acceptor into 4 ml LB broth, respectively. Incubate at 37°C overnight.
  • Transfer 100 μl the overnight culture in step 3 onte the conjugation plate (100 mg/mL sodium azide and 64 mg/mL fosfomycin). Incubate the paltes at 37°C overnight.
  • Any and all advice would be greatly appreciated!
