Dear Hafez, There are many culture collection of bacteria around the world that you have to search. One of them, The American Type Culture Collection (http://www.atcc.org/ ) is the best known. Please, before you get it you have to decide what is the purpose of your research to choose the appropriated culture and also, most important, do you have the appropriate laboratory condition to work with it? Others culture collections are: DSMZ (http://www.dsmz.de/), NCIMB (http://www.ncimb.com/), NRRL(http://nrrl.ncaur.usda.gov/), NTCC (http://www.phe-culturecollections.org.uk/collections/nctc.aspx), JCM (http://www.jcm.riken.jp/) and so on. For my work I went to the local pharmacist, had a look for the bottles of probiotic pills then bought the ones with pure cultures. In Australia we have a local manufacturer by the name of Ethical Nutrients that sells pure and mixed probiotic cultures. Put them up on MRSA to make sure they are LABs. By this method I was able to get pure strains of Lactobacillus rhamnosus GG, Lactobacillus plantarum 299V, and Saccharomyces boulardii. Otherwise you can contact a manufacturer and they will happily give you some isolates. Generally they love to get research done on their products.

Thanks Dr. Latham but when you cultivate these strains did you have to do any additional methods?

Hi Taghreed. All I did was open a capsule, suspend a little of the powder in saline, streak it on growth media (I think it was brain heart infusion agar initially) then incubate at 35oC in 5% CO2. The colonies were all of the same morphology, and consistent with LABs. I checked they were Gram-positive catalase-negative rods, and that was it. If you want to ensure they are the right species (I did not do this) I suggest you do 16S rDNA sequencing with 27F, 1492R primers as shown on Macrogen website. Alternatively contact a probiotic manufacturer and ask for their strains. Does this answer your question?

Hi Taghreed. You can also get probiotics by cultivating yoghurt or other dairy products on MRS medium in 5% CO2 at 35oC for 48-72 hrs.

If you need standard stains of probiotics, i have 3 different ones and you can get them.
