PARP antibody commonly detect full length 116 kda and 89 kda.
I would like to know the specific site in protein sequence where exactly the antibody binding. And is there any specific site for PARP modification such as ADP-ribosylation and acetylation. whether this modification alter the protein activity or not?

As you know PARP is an protein enzyme with average 116 kda MW. It is substrate of Caspase-3 which convert it to p85 averagely. Many companies produce antibody against intact or cleaved PARP (polyclonal & Monoclonal).
If you purchase polyclonal, you can not know about specific site in protein sequence.
You encounter to a mixture antibodies again sequential and conformational epitops.
But about monoclonal antibodies, the producer can help you about the epitop which Ab produce against it.
Some antibody against PARP produce with help of similar synthetic peptides.
e.g. Promega inc produces Anti PARP using a synthetic peptide, gly-val-asp-glu-val-ala-lys (GVDEVAK), representing the N-terminus of the large C-terminal fragment of human PARP.

I want to know specific site for PARP detection. For example 520 or 480, ect...amino acid like that.
I could not get any information like this.
