We have Whatman FTA cards (the cards with a single disc), smeared with dogs faeces within the application spot about 4 months back. we ordered for a QIAamp® DNA Stool Mini spin Kit this week, where we were asked to print the handbook online, I did that. I have tried to extract DNA from the cards using the QIAamp DNA stool Handbook 07/2007 (Protocol: Isolation of DNA from stool for pathogen detection). we have gone through the whole process but on running gel electrophoresis do not have any DNA yield.  Is there any alternative way that we can use to extract DNA from these samples on Whatman FTA cards? I can’t tell if the method I am using is not the right step because I soaked the cut application spot in PBS overnight, vortexed and collected the liquid after removing the pieces of the FTA card suspended overnight. Please, I need your assistance. I look forward to hearing from you.
